Diltiazem coadministration resulted in a significant interaction with simvastatin, probably by inhibiting CYP3A-mediated metabolism
1
Diltiazem: Simvastatin: Diltiazem: Inhibition of CYP3A4 54: Limit dose of simvastatin to no more than 10 mg daily and dose of lovastatin to no more than 20 mg
The interaction of diltiazem and statins, metabolized through cytochrome P450 3A4, leads to increased risk for rhabdomyolysis
Advise people to report any unexplained muscle pain, tenderness or weakness
Diltiazem and atorvastatin combination therapy is also reasonable when necessary, as well as verapamil with lovastatin or simvastatin
Reference: 1) MHRA Drug Safety Update (August 2012)
Verapamil and diltiazem are known inhibitors of CYP3A4; the maximum dose of simvastatin is 20mg daily in patients taking these
2 ± 36
Simvastatin – Diltiazem Interaction Interaction: MAJOR Rationale: MHRA August 2012 does issue do not exceed 20mg simvastatin in combination with diltiazem; Monitoring: Patients should be advised to promptly report any unexplained muscle pain, tenderness or weakness, particularly if accompanied by fever, malaise and/or dark colored urine
Of the total drug interactions, 57 are major, 242 are moderate, and 11 are minor
Subjects received diltiazem 120 mg twice daily for 10 days and on day 10 a single, 80-mg dose of simvastatin was administered

a member of the American Legion, he was the beloved husband of Sandra (Long) diltiazem and simvastatin interaction mhra Davis; loving brother of Richard
Diltiazem (Oral Route) In deciding to use a medicine, the risks of taking the medicine must be weighed against the good it will do
Concomitant use of diltiazem or other potent inhibitors of CYP3A with simvastatin should be avoided, or close clinical monitoring should be used
5)
diltiazem will increase the level or effect of simvastatin by affecting hepatic/intestinal enzyme CYP3A4 metabolism
Simvastatin is used together with a proper diet to treat high cholesterol and triglyceride (fat) levels in the blood
